There is a reason why most clinical labs don’t use LC/MS. If the need to they will use GC/MS for small molecule identification.  They can get much higher throughput and simpler output using current lab techniques. LC/MS isn’t there for clinical lab use for high common lab tests. It’s not simple or fast enough.

Mentions:#LC#MS

For quantification, we see 10% CVs. That's as good as many existing clinical assays. And given that we can look at thousands of proteins at once, and can get additional confirmatory information from each one, I think that can give some pretty reliable results. LC/MS takes 30 minutes for a deep research sample enriched by nanoparticles. A quick and dirty run can be 15 minutes, losing only about 15% of the observable proteins, which is still plenty good enough for clinical purposes.

Mentions:#LC#MS

Overnight isn’t nearly fast enough. I don’t see any validation that the output is sufficiently precise and accurate to apply to clinical assays specifically with regard to quantification.  It just using nanoparticles to extract proteins for analysis with LC/MS. Sure it enriches the samples but the challenges and complexities and limitations of LC/MS remain.  The proteograph just automated sample prep for lc/ms analysis. LC/MS is slow and complex and so is the analysis and reliable quantification is hard That’s why these approaches are used in research labs for biomarker discovery and proteome analysis rather than for clinical applications.

I went from negative net worth to over $1m net worth. Luck has a LOT to do with it. Here's where my path went. 1. Graduated college with a bs in math. Couldn't get a job. 2. Graduated MS applied math. Almost didnt get a job but fought to get a job with a software consulting company. 3. That job was awful but tech was booming at that time so I got a job at a bay area tech company making 6 figures for the first time. 4. Covid happened and dropped housing prices so I bought really low. 5. I refinanced while the fed interest rate was still 0% but my property value had shot up. 6. I gained a special skill that a very desirable company needed so they hired me at $288k. 7. I started investing with a financial advisor. He beats the market by a little bit after his fee and only invests in ETFs. Its not flashy, but its optimal.

There will be no learning. These are people who got where they are in the first place by refusing to learn. They have an outright hatred of the truth. That's why when they lie they don't even do the truth the dignity of crafting a believable lie. Because even that would mean that they acknowledge that people should believe the things they think are true. When they do things like knowingly send an innocent person to a concentration camp, their message is "ha ha ha, our lies are more powerful than your truth. he has MS-13 gang connections or whatever. And Trump, despite laundering money for the Genovese crime family and the Russian Mafia, well he doesn't." If a million+ dead Americans from Covid wasn't a lesson, this won't be either. They want the American Holocaust. They want a soft apocalypse where they think they will emerge as rulers. They are the people who grew up hearing stories about slavery and thought "remember what they took from us." They trust Trump BECAUSE he is a garbage human being who wants to destroy America. If all of this "blows over" they will be PISSED and do everything they can to find another way to bring everything down.

That would make sense...&so would lumping AOC with Sanders now that they're on tour together(my mom of 5, MS brain can make a connection that noone can figure out where it came from 😉) I am tired of the lot of them! While on a fight the oligarchy tour??? The irony is insane. Fact is, they're all the same, remember good ole coach Tommy & his fraud-but he's still a senator & still trading. He's just a snake oil salesman anyway, I lived in AL when he was coach & elected to the senate, he's oily. But so was Mo Brooks, apparently that's how AL likes their politicians-lol. Lindsay Graham is a weasel, his voice irritates me(&its not the southern), however his net worth is small & in 2018 he tried to team up with Warren to stop stock trading(Warren teamed up with Daines in 2022 for the same)....guess Lizzy decided if you can't beat em, join em 🙄 All of them are trash!! McConnell, Feinstein, Burr, Pelosi, Purdue, Loeffler, Gianforte, that entire group(I looked up the 2020 scandal, how sickening???) Ok, I need to stop now. I'm saying any given journalist/news agency gets one shot at it and they would need to agree going in "this guy is getting nailed to the wall" He would leave, of course. My wife is an OT, and I am formerly the coordinator of a University Exercise & Sports Science as well as Athletics department. One of the MAJOR reasons for this huge spike? The ever-escalating war at the mid-tier in every industry (...but especially in Health Care) to require doctoral degrees for licensure. ...and they aren't like traditional doctoral programs. They're more like combined programs that extend the traditional MA course. They rarely involve any kind of thesis research and you don't defend or work as a TA. They're just like...really long capstone projects and extended clinical rotations. My wife took her current job with an MS when the BS was the requirement in 2018. There were, actually, practicing OTs then who actually got grandfathered who had Associates in Science that had then obtained 2 year certificates in OT before it became completely professionalized. As of this year, at her workplace (major state university hospital)...EVERY SINGLE new OT hire must have a doctorate. That will extend every student's time in program by a minimum of 1.5 years and will result, on average, in an additional 30k in student debt. It's almost certain to actually eliminate a lot of non-trad transfers because folks who want to re-train will have exhausted their financial aid eligibility entirely and just won't be able to pay for those programs. Professionalization can be great. And I'm generally a proponent of licensure for all professional programs, but a lot of fields are going to educate themselves right out of existence because students aren't going to be able to afford the time or debt load to complete the degrees in fields where the ROI is like 1.25 instead of 2+

I flipped YT channels between the Brits and US. Overall the Brits sounded smarter not only due to their smarmy accent. The UK journalists had no qualms with hardball questions and the PM had no issues answering them. Trump meanwhile bickers about literal tattoos of MS13 and cries at even softball questions threatening constant press bans lol. The U.K. PM made an obvious but nuanced jab at the US/Trump for acting immature and eroding trust in allies. There was some slight nuance to it so Trump would never figure it out unless Fox News or Laura Loomer translates for him. Basically the U.K. still got a pretty crappy deal framework that was better than lib day but worse than 6 months ago. This is the easiest deal that could possibly be struck logistically. The only reasonable bull thesis is the one MAGA has been selling all along. That the US is exceptional and everyone will need to grovel hand shake and smile for future US preferential treatment deals. That this will end up crippling global economies but JPow should be “smarter” and utilize QE as jet fuel like the rest of the world. This is just about the most flimsy bull thesis I have ever had the privilege of seeing in my entire investing career and if anyone buys into this near their retirement you have my thoughts and prayers.

Looking for advice on an IRA I inherited from my mom's passing. It's a mix of stocks and cash and I'm wondering if I should just sell all of the stocks now and use that to diversify into a few ETFs given the market climate. At a high level: \- My portion that I just received is currently sitting at about 550k (385k stocks / 165k cash) \- The stocks are mostly tech and finance (NVDA, AAPL, AMZN, GOOGL and MS, COF, PRU, BK) I'm not loving the idea of being invested directly in individual stocks even though the account has done well over time; not very optimistic about the next few months in the markets. That being said my initial thoughts were to sell all the stocks now, and then use the funds to purchase VTI and VXUS. If I decide to do this, what would you say is a reasonable time range to cost average into the two etfs? Should I buy some each week over the next year until all of the funds are in the two etfs? Go longer or shorter? I don't want to wait too long to get the money in but again, being cautious about the next few months makes me think it would be worth it to do it over at least the year. One last bit of info, I can only really take the money out of the IRA over the last 2-3 years of the required 10 year withdrawal period due to tax concerns, that being even a small withdrawal would more than likely put me in the 37% tax bracket, so I plan to take the funds out when I am no longer working in year 8/9/10, not sure if that impacts any suggestion on strategy. Thanks for the opinions in advance!

Search is \~60% of Google's revenue. If you do not think AI search is going to supplant regular search, then yes, Google is on sale currently. I'm not sure if I've really used Google since Deep Research and o3 have come out. So on that front, if you do think AI search is going wi (I do), then it's a lot more complicated. Google would have to grow the other 40% share of its revenue fast enough for the 60% going into free fall to not matter. They have to do this while maintaining total revenue, or at least, margin (mass layoffs, selling buildings/infra, etc -- that's how most companies in similar positions try to approach it. Deep, deep cuts) Or they have to have a story that's so exciting that enough people will apply a huge premium to justify paying a higher speculative price. And they have to do it as a \~30 year old company with a pre-existing \~2T valuation (tons of risk for not a lot of upside. 2x at most.) Now suppose the market (and common sense) thinks we're heading into a recession. In that case, YT and Waymo revenue will both have downward pressure for the foreseeable future (advertising gets rocked in down turns, people go to dinner/drinks/shopping/travel less, etc.). That only complicates their chances because the revenue will be difficult to grow. Now let's compare that to Apple and MS. AI isn't (yet) threatening the iPhone, and Bing is a rounding error for Microsoft. Apple revenue depends on the iPhone and could face a threat there if one were to emerge, but MS's revenue is much more diversified. Azure is their leader, and it's \~40% of their revenue, not a majority. Their other 60%, AI probably stands to enhance (i.e. Excel becoming more data science oriented, AI at an OS level, etc) Google's challenges aren't insurmountable though. If search were 70%+ of revenue, it might be, but 55, 60%? Maybe. It's just a big maybe, and recession pressure complicates it. So does Google being a bit infamous in tech for bureaucracy, and the fact that they haven't done this before, or even really innovated internally in a long time (most cool shit has been bought -- yt, waymo, android, etc). Just seems to me like the same story we've seen a dozen times in tech — managed decline of a once dominate giant. There's better and more exciting choices with less risk and more upside.

You are misremembering. Period 1, oh shit things are getting squirrelly and that was mid to end of 2007. Bear hedge fund fails, BNP starts blocking withdrawals from their hedge funds, and then we go into a recession. Beginning 2008 is where the real fun starts. Fed cuts rates 2x in 10 days in January, Bear is a fire sale , Freddie and Fannie are nationalized, then Lehman fails, AIG fails(too big to fail). September 2008, Hank Paulson comes up with TARP. WaMu fails. Beginning of October Dow falls 20% in a week. November 2008 the Fed starts buying everything. There was a time during 2008 you did not know if Citigroup, JPM, Wells, MS would even exist the next day. Many large, profitable companies were down 80-90%. For example look at a 20 year chart of Citigroup (C)

I finally just watched the interview 🥭did about the photoshopped picture of the MS13 on the guys knuckles. Wow, talk about hilarious. It reminds me of the crazy drunk guy at a party trying desperately to explain something to the sober guy. “Wait, wait, you’re not listening. He had M S 1 3 on his hand”, Shhh, you’re still not listening.” Lmfao.

> Build an AWS or Azure from scratch? https://invidis.com/news/2024/03/it-security-china-blocks-use-of-intel-amd-and-windows/ Funny how China can survive and grow extremely fast whithout relying on AWS, Azure etc... The thing is, all the ***needed*** building blocks for datacenters are available. Open Stack is successfully used by many companies. It's an effort, but we don't need to rebuild all the managed services AWS offers to create the same level of vendor-lockin. > 80 billion a year on AI and Datacenter costs. Yeah, we will see where the whole AI thing goes... The problem with AI is, it's not the sum of global intelligence and knowledge, it's the average. Only much faster. > not unreliable renewables Boy, put the cool-aid aside 🤣 That's what energy-storages are for and why we invest in them. A portfolio of renewables is quite reliable with some buffer in between. > We don’t think about you at all. Hahahaha yeah, sure. Thats why MS started all the whitewashing, chaning their legal team, to disguise themselves as Trump-critics to the outside market. Because they aren't thinking about us ***at all***.

Although this sentiment is an absolute mischaracterization, the fact that Trump needs to surround himself with ass kissing rubes is the weakest, most pathetic administration we’ve ever had. Why do you think he’s characterized as a crying orange baby in diapers? Because that’s how the world sees him. And he’s either losing his mind or dumb AF, “MS13 on his knuckles”!

I think by the time he (or the people around him) realize that they are in big trouble in regards to the midterms, it's going to be way too late. Supply chains are going to be broken, small businesses too far under-water to recover and consumer confidence out the window. He is crazy obstinate (see MS13 tattoos) and the only possible exit from this train wreck is a sudden fatal health emergency.

The market is always in a constant flux and in 5 or 10 years while we may remember issues we faced, the top companies and who the major players are will different. * Jan 2000 - Cisco, IBM, and Oracle were three of the top 5 stocks by market cap. * Jan 2010 - Apple and Google showed up in the top 5. * Jan 2020 - Amazon and Facebook became the biggies, Tesla as well but they hadn't reached top 5 at this point. * Jan 2025 - Nvidia is the new one in the top 5 * The only company that has been in the top 5 by market cap during each of these periods was MS. My point? Your fav company or your fav influence on the economy is going to end and go away, the only truth is what the market actually ends up doing, so I dunno, take whatever advantage you can, get your gains, cut your losses, do some blow off a strippers ass or her lady D when you can and enjoy it.

Between this, and his insistence that that dude had MS-13 tattooed on his knuckles I'm beginning to believe that Donald Trump has absolutely no idea what's really going on or what his administration is actually doing. It seems apparent that he has surrounded himself with sycophants who only praise him for every decision or move that he makes. Generally they blow smoke up his ass and do whatever the fuck they want, and he simply signs whatever they put in front of him and doesn't question any explanation that they give him. Everyone always trying to give Donald Trump credit for the atrocities of his administration... When the reality has always been that he is nothing more than a popular mouthpiece being stood up to attract higher while all of the real atrocities are being done by other people out of sight. I really don't think that he has any idea what the fuck is really going on.

Yeah the writing is on the wall. The future of PCs is going to be ARM versus x86. Apple already made the transition. MS is slowly getting there but with more powerful chips coming from Qualcomm that transition will be easier to push. I love my M series Mac and iPad products. Nothing on the market compared to the performance and battery life right now. The surface pro with Qualcomm chip is good but still not as good as what apple offers. And they have nothing to compete with the M4 Max chips yet.

Microsoft can fight it in court if it wants, but the federal government has broad authority to regulate international commerce. This issue has already been adjudicated several times, and well-established case law exists. They are, of course, free to try to fight..... and lose... The President could unilaterally sanction MS's overseas operations, and there is nothing they can do about it. The President could also unilaterally tariff MS's export of services at 1,000,000,000,000% and there's nothing they can do about that either. But it's all moot, because none if that shit is happening.

I transferred my MS account over to Schwab. Schwab took care of it. Without advisors you will be able to manage the accounts.

Mentions:#MS

Yeah... well, funny you mention it. My mother called "their guy" and asked him to call me to discuss the holdings. He naturally has a vested interest in keeping the account with MS as opposed to Fidelity/Schwab/Vanguard. He warned me that my parents would pay $50,000 in taxes if they sold everything (currently \~$500K in it; no idea what the basis for the stocks are so I can't confirm this tax guesstimate). Which is true, of course, but he insisted the portfolio is "doing well" for them and then asked what other assets they had that could be transferred to MS. Felt very sales-ish. Trying not to give up too much information here, but suffice to say the guy is a very old friend of my father's so I tried to stay in my lane; it's not my money after all. But knowing the condition of my parents' age/health and then making efforts to get more under MS management was disappointing. At the end of the day, nothing is on fire here. She doesn't need the money in the brokerage account and uses the dividends to pay IRS quarterly estimates. It just bothers me that she can't take action on the account by herself and will have to talk to "their guy" who will likely dissuade her from touching anything.

He's probably difficult to communicate with. You see that disaster of an interview where he demanded Terry Moran "admit" that Kilmar Garcia had "MS13" tattooed on his hand? He clearly just didn't know that the characters were photoshopped likely by someone in his own administration and excused as "interpreting" the symbols on Garcia's hand (which just meant "smoke weed to get high; pray to god until you die"). Ignoring that the MS13 is so obviously photographed that not even a modern elementary schooler should be fooled, how has no one on his team communicated these facts to him as they gave him that photo of the tattoo? Yes, they're wrong about what the tattoos meant, but they could have at least told him that they photoshopped characters were interpretations, not actual tattoos.

Got AMD when it tanked. Was like $1-1.50/share. All the stock news was saying it was done. All the tech nerds were crapping on it vs intel. But, others news was coming out saying both MS and Sony were tapping AMD to create APU's for their next-gen consoles. Big companies aren't tapping someone to do something big for them if the contract company is going out of business. To me, it sounded like a win-win; AMD got some big contracts, and MS/Sony got to probably cut some sweet deals for those APU's. I ended up going back to college right as the stock started going up. I had to sell all my stuff to qualify for student loans. Sold the stock for $3000 and paid for a cruise for me and my wife. First vacation she had after she graduated college. If I had kept the shares, I think I calculated they would have been worth like $125k or something. Not exactly "millionaire maker" territory. (I was working a shit job and didn't have much to invest). But, being a broke-ass college student and being able to take my wife on a cruise was priceless.
